Epitalon

A four amino acid peptide with striking longevity claims resting almost entirely on one Russian research group, and essentially no independent replication.

What it is
Epitalon is a synthetic tetrapeptide, alanine-glutamate-aspartate-glycine, developed from an extract of the pineal gland by Vladimir Khavinson's group in St Petersburg. It is meant to be the active fragment of that extract.
The claims attached to it are the largest on this site: telomerase activation, restored melatonin rhythm, extended lifespan.
Why the evidence needs care
The core work comes almost entirely from one research group over several decades. There are cell studies reporting telomerase activation in somatic cells, rodent work reporting increased lifespan, and long-running human cohort studies reporting reduced mortality in elderly subjects.
That last category sounds decisive and is the weakest. The human studies are largely unblinded, conducted by the group that developed the compound, published in a narrow set of journals, and have not been independently replicated outside that lineage. None of that proves they are wrong. It does mean the evidence has never survived the test that matters, which is somebody else trying to reproduce it and succeeding.
For comparison, BPC-157 sits in the same tier with over a hundred rodent papers from multiple groups. Epitalon has fewer papers from fewer independent sources, and considerably bolder conclusions.
The telomerase question cuts both ways
Telomerase extends telomeres, and short telomeres are associated with cellular ageing. That is the appeal.
It is also, precisely, one of the mechanisms by which cancer cells achieve unlimited replication. Most human cancers reactivate telomerase to do it. A compound whose headline mechanism is switching that enzyme back on in somatic tissue deserves more caution than it usually receives, and nobody has run the long-term human study that would settle it.
Practical notes
- The cycled protocol is inherited, not derived. Ten to twenty day courses come from the original Russian regimens; there is no dose-finding work behind the numbers.
- Sleep effects are the most consistent self-report. Given the pineal origin and the melatonin claims, that is at least mechanistically coherent, and it is also the sort of thing that responds well to placebo.
- Treat lifespan claims as unproven. Not disproven, unproven. Nothing on this site has a wider gap between what is asserted and what has been independently shown.
Side effects and warning signs
Commonly reported
- Injection-site irritation
- Occasionally reported drowsiness after dosing
- No systematic human safety dataset exists, so the absence of reported effects reflects absence of study
Stop and get medical help
- Any active malignancy. Telomerase activation is the mechanism claimed for this peptide, and it is also one of the things cancer cells do to become immortal
- Unexpected systemic reaction, since there is no established baseline for what normal looks like here
